About the Founders:

William Sellers is a core institute member of the Broad Institute of MIT and Harvard, a faculty member at Harvard Medical School, and faculty member and senior advisor to the president for experimental therapeutics at the Dana-Farber Cancer Institute.
Eric Fischer is an Assistant Professor in the Department of Biological Chemistry and Molecular Pharmacology at Harvard Medical School and an Independent Investigator, Dana-Farber Cancer Institute.


In this role, part of the Lead Discovery team at Civetta, you will work under the supervision of a Senior Scientist to develop and implement a pipeline for expression of human proteins in both bacterial and insect cell systems. Your skills in molecular biology and cell culture will be critical to experiments designed to provide protein reagents for downstream work in high-throughput screening and structural biology at Civetta. This is an excellent opportunity to learn and grow in the unique environment of a start-up biotech in Cambridge.


Key Responsibilities:

  • Assist in designing and creating expression vectors for use in bacterial and insect cell heterologous expression systems using robotic liquid handling systems
  • Perform small-scale and pilot-scale expression trials to identify conditions for soluble expression of human proteins in heterologous systems using robotic liquid handling systems
  • Scale-up to liter volumes of bacteria in high-density bio-fermentation systems, and of insect cells in shaking incubators


Required Skills and Qualifications:

  • MS or BS degree in biochemistry, molecular/cell biology, or equivalent with 1-2 years of laboratory experience
  • Solid experience with standard molecular biology techniques including primer design, PCR, and subcloning into expression vectors desired. Exposure to high-throughput use of these technologies (96-well or 384-well PCR, colony PCR screening, ligation-independent cloning) preferred, but not required.
  • Experience in bacterial protein expression systems required; experience in insect cell expression systems valuable but not required
  • Exposure to protein biochemistry techniques in small scale, for example analysis of soluble expression using Western blotting and/or small-scale purification of protein utilizing affinity resins, would be optimal.
  • Familiarity with Opentrons or other robotic liquid handling systems, or willingness to learn these tools, a strong plus
  • Good attention to detail, and excellent time management, to enable use of electronic data notebooks and database software in a timely manner to track/annotate plasmid and protein stocks
  • Pride in performing high-quality, reproducible results, and willingness to communicate weekly and monthly scientific progress to team members on an informal basis
  • Eager to work on tight deadlines and troubleshoot/optimize protocols within those deadlines to deliver quality reagents to internal stakeholders
